PhotonicSweden Optopub at HÜBNER Photonics

On the 16th of February 2023 we hosted the years first Optopub with Photonics Sweden where we combined the event with a soft opening of our new office expansion.

Real-time single-molecule 3D tracking in E.Coli

Presented by Elias Amselem, Department of Cell and Molecular Biology, Uppsala University

We have little detailed knowledge about the intracellular dynamics of fundamental processes such as gene regulation or protein translation; the intracellular environment is very different from the dilute conditions where most of what we know about biochemistry has been learned. Here, we present a 3D single-molecule tracking system based on cross ¼ entropy. Putting the system into practice, we track single fluorophores within the Escherichia coli bacterium. A fluorescent marker is attached to the E. coli trigger factor (TF), a bacterial chaperone protein that binds to translating ribosomes close to the peptide exit tunnel. The system can locate a reporter in a volume of size 0.95×0.95×1.40 of 0.84 ms at a photon count rate of 120kHz 

Transient State (TRAST) Spectrocscopy and Imaging of Cellular and Molecular States and Conditions – Exploiting the Sensing Side of Fluorophore Blinking Kinetics

Dark state transitions of fluorophores are central for all forms of fluorescence-based, single-molecule and super-resolution microscopy and spectroscopy, mainly as limiting factors in single molecule studies, at the same time prerequisites for super-resolution imaging techniques. This overview will highlight an additional aspect of such transitions, namely that they can be used to sense a manifold of biomolecular environments, dynamics and interactions. Principles for how they can be imaged will be described, as well as application examples in cells and tissues.

The B-Brighter Project

The B-BRIGHTER project is an EU-financed research project with the goal of developing a novel, light-based bioprinting technology. The project utilises continuous-wave lasers, acousto-optics, galvanomirrors and custom hydrogel-based bioinks to make this concept a reality.

Benchmark evaluation of a single frequency continuous-wave OPO seeded pulsed dye amplifier for high resolution laser spectroscopy

For short-lived radioisotopes, it is of special interest to study their nuclear structure. Production of single isotopes is strongly dependent on the ionization method. One uses multiple laser beams, each precisely tuned to match successive unique electronic transitions. For high-resolution spectroscopy, narrow pulsed laser linewidths are created by the pulsed amplification of a narrow-band cw laser, which takes the amplifier’s power and time profile whilst acquiring the seeder’s spectral properties. This work demonstrates a pulsed dye amplifier seeded by a CW C-WAVE OPO as the first excitation step for high-resolution spectroscopy of radioactive silver in the CRIS experiment at ISOLDE, CERN.

Optimization of higher harmonic generation microscopy for acute tissue imaging

For patients suffering from cancer or neurodegenerative diseases research into the underlying causes is of the highest importance. Here, we report on our efforts to optimize higher harmonic generation microscopy complemented with multi-photon (auto)fluorescence, to image for long period of times on acute human tissue slices. Results for time-lapse imaging of the induction of myelin-swelling in Multiple Sclerosis brains during gradual high sodium perfusion and fibrosis induced in samples obtained from lobectomy, in the context of pulmonary fibrosis will be presented.

What is Day of Photonics? and why do we celebrate it?

On 21 October 1983, the General Conference of Weights and Measures adopted the value of 299,792.458 km/s for the speed of light.

In honor of this occasion, we celebrate the International Day of Photonics on October 21st every year. This year we at HÜBNER Photonics are very excited to be hosting a photonics and lasers inspiration evening for students at KTH Royal Institute of Technology in Stockholm, the 17th of November. We hope we can ignite sparks of inspiration for future researchers, as we continue our journey into the photonics industry of tomorrow.

Fluorescence imaging using lasers is a key technique in both biomedical research and clinical applications. Fluorescence-based microscopy for high-resolution and high-throughput multifluorophore imaging typically relies on the use of several individual laser sources at different wavelengths. One option to achieve this is a permanently aligned and service-free multicolor illumination source. Such a multi-line laser solution, with up to four laser wavelengths in a small package, can enable more cost-efficient, compact, and user-friendly instrumentation, in addition to simplifying manufacture and maintenance.

Haahr discusses the relative advantages of multi-line lasers, introduces the technology, and illustrates a real-life example in which multi-line lasers are helping to simplify system design and manufacture for instrumentation, which can, in theory, address the speeds necessary for clinical sampling. The session is intended for both researchers and instrument manufacturers who are interested in new solutions of lasers for their experiments or instrument design.

News from SCiX 2022

Thanks to Andor for presenting their informative poster at SCiX 2022, with a mention of our Cobolt Tor XE and how it helped acquire the ground breaking results using LIBS!

Our Amanda Amori Faltyn stopped by to chat with the author, hearing more about the exciting results from Proff. Motto-Ros and his team at Université Claude Bernard Lyon 1. The results show, for the very first time, high resolution, 4K elemental distributions of geological samples obtained using uLIBS with an acquisition time of only 3 hours. Our Cobolt Tor XE pulsed laser was used together with the fast gated sCMOS camera from Andor Technology to generate this spectacular data. Very exciting! #LIBS #lasers #pulsedlasers #scix2022
